Twinklestar Sprites is a 2D vertical scrolling shooter that also integrates object-dropping puzzle elements. The screen is divided vertically into two halves, with one player taking a left half and the CPU or second player taking the right half. Each player controls some type of flying unit that can shoot both single shots and a charged super shot. Units can also throw two screen-clearing bombs per round.
The object of the game is different from most titles in the genre. Instead of progressing through stages by defeating bosses, players have to outlast each other. Each player has a row of filled hearts indicating his or her flying unit's current health, and when all those hearts are gone from taking hits from enemies, the unit is destroyed and the player loses that round.
Twinklestar Sprites also has a head-to-head element, in that whenever players destroy enemy units in a chain combo, they're sent to the other player's screen in certain patterns. By destroying many ships at once, players can clutter up their opponents' half of the screen, making it more difficult for them to survive.
There are three modes for players to choose from. Character Mode lets players select one of the game's nine characters, each rated differently in speed and shot, to take on all the other characters. In Story Mode players can only control the game's main character, but the game progresses with story scenes before and after matches.
